How does Protandim Work?
The big question is – why does the synergistic blend of herbs in Protandim work to produce more antioxidant enzymes that prevent cellular damage by free radicals?

“This is an incredible breakthrough. Prior to Protandim, science was unable to find a way to get the cell to produce more of the antioxidant enzymes that prevent cellular damage by free radicals. As I said, scientists have known for years that dietary antioxidants can’t do the job that must be done. Concurrently, research was telling scientists that oxidative stress is extremely damaging as it came up over and over again as being associated with the onset and/or development of diseases. Even scientists studying aging knew that free radical damage to the cells was a significant contributor to the process we call aging.
Scientists knew what the problem was and where the answer had to be, but they had no idea what method to use in order to get the DNA to make more of the cytoprotective enzymes.
Dr. Joe McCord and his fellow researchers began experimenting with all-natural ingredients, used for literally centuries in Ayurvedic and Chinese medicine. What they found, and eventually Patented (there are now three Patents on Protandim) was a particular formula, a recipe, that produced an 1800% (18 times) larger increase in cytoprotective enzyme production than any single ingredient, or any other combination of ingredients. Nrf2 Goes Beyond Production of Antioxidant Enzymes
Nrf2 does far more, it turns out, than what Dr. McCord and his team had first envisioned. They simply wanted the cell to produce more SOD, catalase, and glutathione, but their invention clearly did more than that. The following is from a research paper published in November of 2010 in the scientific journal Integrative and Comparative Biology by researchers from the Department of Cellular and Structural Biology at the University of Texas (read just the italicized part if you don’t want to wander into the complex world of biochemistry too deeply):
Nrf2, a guardian of healthspan and gatekeeper of species longevity:
Although aging is a ubiquitous process that prevails in all organisms, the mechanisms governing both the rate of decline in functionality and the age of onset remain elusive. A profound constitutively upregulated cytoprotective response is commonly observed in naturally long-lived species and experimental models of extensions to lifespan (e.g., genetically-altered and/or experimentally manipulated organisms), as indicated by enhanced resistance to stress and upregulated downstream components of the cytoprotective nuclear factor erythroid 2-related factor 2 (Nrf2)-signaling pathway. The transcription factor Nrf2 is constitutively expressed in all tissues, although levels may vary among organs, with the key detoxification organs (kidney and liver) exhibiting highest levels. Nrf2 may be further induced by cellular stressors including endogenous reactive-oxygen species or exogenous electrophiles. The Nrf2-signaling pathway mediates multiple avenues of cytoprotection by activating the transcription of more than 200 genes that are crucial in the metabolism of drugs and toxins, protection against oxidative stress and inflammation, as well as playing an integral role in stability of proteins and in the removal of damaged proteins via proteasomal degradation or autophagy. Nrf2 interacts with other important cell regulators such as tumor suppressor protein 53 (p53) and nuclear factor-kappa beta (NF-κB) and through their combined interactions is the guardian of healthspan, protecting against many age-related diseases including cancer and neurodegeneration. We hypothesize that this signaling pathway plays a critical role in the determination of species longevity and that this pathway may indeed be the master regulator of the aging process. These researchers clearly did not know about Protandim, or they would have known that a method had been discovered that activated Nrf2 without having to induce cellular stressors (I will explain this below). However, they do understand Nrf2. Look at the portion of the paper I’ve highlighted. It would seem that Nrf2 is extremely good for cellular health, which means it’s good for your overall health.
Other researchers, including Dr. McCord, now say that there are far more than 200 genes affected by Nrf2, citing numbers as high as 500 genes (these genes are often referred to as the “survival genes” because they provide the products needed for cellular health and cell survival). This is cutting edge science and the scientists are learning more as research continues. What is clear, though, is that Nrf2 is the key to healthy cells.
Dr. McCord gave a wonderful lecture on what Nrf2 does, which he called, “Tuning the Genome.” The analogy he used was that, as we age, as we go through various traumas to our bodies and minds and as we simply accumulate more years being alive, the genome is like an old piano—it works, but it is in desperate need of tuning. Certain notes (the production of certain enzymes like the antioxidant enzymes) that should be high are now low. Other notes that should be low (like the production of inflammatory proteins) are far too high. Nrf2 “tunes the genome” by putting products of the genome back to where they should be, just like a piano tuner puts the frequency of each string in the piano back to where it should be — where it was when the piano was new.
It seems to me that there is really nothing you could do that is healthier for your body at the cellular level than taking Protandim and activating Nrf2.
Explanation of the “ROS Pathway”—Inducing Nrf2 Through Cellular Stress:
In the paper on Nrf2, cited above, they mention the reactive oxygen species pathway to inducing Nrf2. Reactive Oxygen Species (ROS) are free radicals. The ROS pathway is basically inducing a condition where there is such an enormous overload of free radicals that the cell reacts by releasing Nrf2. As we’ve seen, this is something the body is increasingly reluctant to do as we age.
One of the things we see in elite endurance athletes is that they are so overloaded with free radicals (due to high metabolism and unusually high intake of oxygen) that their bodies actually produce more Nrf2, and therefore they have higher concentrations of the antioxidant enzymes than normal people.
Here’s the question, though: How much damage do you want to do before taking care of the problem? It’s not like the damage to the parts of the cell caused by the increased oxidative stress–prior to the antioxidant enzymes kicking in–goes away. Wouldn’t it be better to prevent the condition than have it get so bad that the body is finally forced to take action?
There are products on the market that claim that they use the ROS pathway (rather than the Nrf2 pathway that Protandim uses) to induce production of antioxidant enzymes. One way this is done is by formulating a drink that contains free radicals, like hydrogen peroxide and chlorine (in forms tolerable to the human body). The idea is to build up so much oxidative stress that the cell will defend itself. Again, how much damage do you want to do before you get protection?
Here’s an analogy comparing use of the ROS pathway and Protandim’s method for inducing
Nrf2 and the protective antioxidant enzymes:
Picture your cell as a nightclub, and Nrf2 is the floor manager in charge of security. In the back there is a team of bouncers (the antioxidant enzymes) that just hang out until called upon to bounce unruly patrons from the club. Protandim is like having a cop walk in as the club starts to become crowded and telling the floor manager that he’d better get the bouncers out onto the floor because a large and rough crowd is on its way. The floor manager gets the bouncers out and they control the crowd, not allowing damage to take place. The ROS pathway works differently. The large and rough crowd comes in without warning. It gets more and more out of hand and begins overturning tables, breaking bottles, fights break out, the bartender is beaten up, and the mirror behind the bar gets broken… Finally, and belatedly, the floor manager gets fed up and he makes his way through the rubble to get the bouncers who clean out the troublemakers, but the damage is already done.

Joe Milton McCord (March 3, 1945- ) is an American biochemist and current Professor of Medicine at the University of Colorado at Denver. He is most famous for his discovery, along with Irwin Fridovich, of the enzyme superoxide dismutase. For this work, Dr. McCord has earned numerous awards and recognition. He has been a member of the board of directors of the LifeVantage Corporation (makers of the dietary supplement Protandim) since 2006.
Dr. McCord hold a B.S. in Chemistry from Rhodes College (graduated 1966) and a Ph.D. in Biochemistry from Duke University (graduated 1970), where he also conducted postdoctoral research.
Dr. McCord has previously held biochemistry faculty positions at Duke University Medical Center and University of South Alabama. He currently serves as a professor in the department of Biochemistry, Biophysics, and Molecular Genetics at the University of Colorado Denver.
Dr. McCord has received several awards and honors for his research and discoveries in the field of free radical biochemistry. McCord is a past recipient of the Elliott Cresson Medal, the Discovery Award from the Society for Free Radical Biology and Medicine (SFRBM), and a Lifetime Achievement Award from the Oxygen Society. He holds an Honorary Doctorate from the University of Buenos Aires and has served as Honorary President of the International Society of Antioxidants in Nutrition and Health (ISANH) and as Chairman of the Third International Conference on Superoxide Dismutases at the Institut Pasteur.
